PLH Genetics
Review & explanation report
- Patient: 4-year-old boy (anonymized)
- Test: clinical exome
- Date: Sep 12, 2026
1In short
Your son's test found a change in the GAA gene, which is related to Pompe disease. With today's evidence, the lab can't say whether this change causes the disease. That's why it calls it a variant of uncertain significance (VUS). This is not a diagnosis.
2What the test looked at
The exome checks the part of the genes that holds the instructions to make proteins. That's where most changes that explain rare diseases are found. It doesn't check all of the DNA.
3What the result means
An uncertain variant shouldn't be treated as the cause. Over time, many are reclassified, most often as harmless. What matters now is whether other data (symptoms, tests, family) can help clarify it.
4Questions for your next appointment
- Do my son's symptoms match Pompe disease?
- Is it worth measuring GAA enzyme activity?
- Should we test the parents?
Recommendation
In this case, an in-depth reanalysis of the variant could bring additional clarity.
